研究组工作摘要:
在本年度我实验室的工作主要集中在1).人源化细胞外基质的研究,我们已取得了全人源化的细胞外基质,已初步验证其可支持人胚胎干细胞的生长。2).利用BAC
clone进行特意细胞组织的标记。 我们已初步建立了BAC同源重组的方法,标记了OCT-4基因。我们将利用此方法进一步标记BRACHYARY;NKX2.5等与心肌细胞发育有关的基因,以便研究TGF-BETA,WNT家族在从人胚胎干细胞到心肌细胞分化过程中的功能。3).人胚胎干细胞的自我复制与成体细胞的去分化是干细胞研究的重大研究方向之一,我们实验室正在建立一套快捷的评估方法,以便进行快速筛选与人胚胎细胞自我复制与成体细胞的去分化相关的基因。
Research Description:
In 1998, Dr. James Thomson in Wisconsin Regional Primate Research Center, University of Wisconsin derived human stem cells and established the first human stem cell line, which is a significant event in regenerative medicine. One important direction in regenerative medicine research is to transplant cardiac myocytes differentiated from human embryonic stem cells to treat myocardial infarction.
Overall, our research focuses on:
◆Investigate the mechanism of the differentiation of human embryonic stem cells to cardiac myocytes using molecular, cellular and genetic methods, which will help to facilitate the treatment of myocardial infarction using cell transplant method.
◆Investigate the function of extracellular matrix in the self-regeneration and directional differentiation of human embryonic stem cells using biochemical, molecular and cellular method.
